History
Advaxis’s official date of inception was 1 March 2002. Its corporate history was closely connected to a public-shell transaction completed in 2004. Great Expectations, a Colorado corporation incorporated in 1987, acquired Advaxis in November 2004. The acquiring company changed its name to Advaxis in December of that year and later reincorporated in Delaware. This structure helped establish the corporate vehicle through which the biotechnology business pursued financing, research, and public-market development. The company’s scientific identity came from a platform based on live but attenuated Listeria monocytogenes. The vector was genetically engineered to secrete a fusion protein containing a selected antigen and an adjuvant associated with Listeria’s immune-stimulating properties. Advaxis described the approach as an Lm-LLO immunotherapy platform. Programs addressed additional oncology settings, including breast cancer and personalized immunotherapies based on individual tumor antigens. The company also investigated applications outside human oncology. Under a cooperative research and development arrangement with the Department of Homeland Security, a pilot proof-of-concept study examined safety in cattle. Separately, Advaxis and the University of Pennsylvania School of Veterinary Medicine began a Phase 1 study of an Lm-LLO-based immunotherapy in canine osteosarcoma. Commercial development was pursued through partnerships as well as internal clinical work. In 2014, Advaxis entered a co-development and commercialization agreement with India’s Biocon for ADXS-HPV in the Indian market. The arrangement was relevant to HPV-associated cancers, particularly cervical cancer, but the company’s programs remained investigational and did not become a broad portfolio of approved commercial medicines. Advaxis subsequently underwent corporate consolidation. A merger with Ayala Pharmaceuticals led to the combined company beginning operations under the Ayala Pharmaceuticals name in January 2023.
